When the wee smily face appears in the chat column of uploads pane, does that mean the person wants to chat with me? Or is it that have simply enabled chat in general?

It simply means they have Chat enabled. It's much easier to Direct Connect & Browse people who do have Chat enabled. Particularly those uploading from you (see Monitor window.) You'll know when you do receive a message. See below:
