Check with your isp. RoadRunner is known to filter p2p apps.

port forward the port's for the program and if your a 64 bit proccessor user use the sp2 patch also make sure the program is vista compatable and if not set the compatability mode to xp and try. may even want to use the xp compatability mode anyway's. right click program icon and click properties, select compatability mode and choose xp.
